Effect of different plant materials on vegetative development of Lentinus edodes (Shiitake) edible mushroom

Öz

Lentinus edodes micelles (LEM), an edible cultivated mushroom, were investigated. In accordance with the purpose of the study; Maize Flour (MF), Rice Flour (RF) and Wheat Flour (WF) with 20%, 15%, 10% and 5% concentrations, MF, RF, WF, mixture and malt extract were used. The effect of different media on mycelial Development Time (MDT) has been shown. Mulberry Sawdust (MS), Poplar Sawdust (PS), Oak Sawdust (OS) and a mixture of these materials were used as ligno-cellulosic material in the seed mycelium and compost medium. Rice Bran (RB) and Wheat Bran (WB) were used as nitrogen source (22% bran:78% sawdust). The data obtained with the SPSS 17.0 statistical program were evaluated statistically. It was determined in the shortest 5% RF medium with 12.4 days of MGS, and the longest in Petri dishes with 20% WF with 17.4 days of MGS. In the production of seed mycelium; It was observed in the shortest mixture with MGT of 19.2 days, and the longest MS with MGS of 22.8 days. For the culture medium, 18 days of MGT and the shortest MS were seen in the medium with RB, and the longest with MGT of 36.8 days was seen in the medium without additives. In this study, the most suitable medium for the vegetative growth of Lentinus edodes edible fungus in the laboratory environment, 5% RF agar medium; It was concluded that the mixture medium for the production of seed mycelium (spawn) and the culture medium containing 78% MS and 22% RB for compost production are the most suitable media.

Lentinus edodes (Shiitake) yenilebilir mantarının vejetatif gelişmesi üzerine farklı bitkisel materyallerin etkisi

Öz

Mevcut çalışma ile yenilebilir bir kültür mantarı olan Lentinus edodes misellerinin (LEM) vejetatif gelişmesi üzerine farklı ligno-selülozik maddelerin etkileri incelenmiştir. Bu amaçla; Mısır Unu (MU), Pirinç Unu (PU) ve Buğday Ununun (BU), %20, %15, %10 ve %5’lik konsantrasyonları ile MU, PU, BU karışımı ve malt ekstrakt kullanıldı. Misel Gelişim Süresi (MGS) üzerine farklı besi yerlerinin etkisi gösterilmiştir. Tohumluk misel ve kompost ortamındaki ligno-selülozik madde olarak, Dut Talaşı (DT), Kavak Talaşı (KT), Meşe Talaşı (MT) ve bu materyallerin karışımı kullanıldı. Pirinç Kepeği (PK) ve Buğday Kepeği (BK) azot kaynağı olarak kullanıldı (%22 kepek:%78 talaş). SPSS 17.0 istatistik programı ile elde edilen veriler değerlendirildi. 12.4 günlük MGS ile en kısa %5’lik PU ortamında, 17.4 günlük MGS ile en uzun %20’lik BU’lu petrilerde belirlendi. Tohumluk misel eldesinde; 19.2 günlük MGS ile en kısa karışımda, 22.8 günlük MGS ile en uzun DT’de gözlendi. Kültür ortamı için 18 günlük MGS ile en kısa DT’nin PK’li ortamında, 36.8 günlük MGS ile en uzun KT’nin katkı maddesiz ortamında görüldü. Bu çalışma ile Lentinus edodes yenilebilir mantarının laboratuvar ortamındaki vejetatif gelişimi için en uygun besi yerinin, %5’lik PU’lu agar ortamının; tohumluk misel (spawn) eldesi için karışım ortamının ve kompost üretiminde ise %78 DT ile %22 PK içeren kültür ortamının en uygun ortamlar olduğu sonucuna varılmıştır.
